For the 2012 Lamar Hunt US Open Cup, U.S. has publicized the probable pairings and venues for the quarterfinals. The games of the fourth round will be held on Tuesday, June 5. The result of that round will decide the teams and combinations for the quarterfinals going to be held on Tuesday, June 26.

Among the sixteen left over sides, eight first-division clubs are from Major League Soccer, three second-division teams are from the North American Soccer League, three third-division teams come from USL Pro, and the rest two teams come from USASA (amateur) provincial qualifier and Premier Development League (amateur).

The current tournament is the ninety-ninth edition of the US Open Cup. The management of the current edition has applied a random picking method for deciding home sides from the primary round to the quarterfinal matches. The teams are determined by coin flip, which has given partakers an equal opportunity for hosting their potential challengers for these stages.
